IZB-Alumnus MorphoSys AG (FWB: MOR; NASDAQ: MOR) gab Mitte September bekannt, dass die US-amerikanische Gesundheitsbehörde FDA (Food and Drug Administration) für Tulmimetostat, den dualen EZH2- und EZH1-Inhibitor der nächsten Generation, den Fast Track-Status für die Behandlung von Patientinnen mit fortgeschrittenem, rezidivierendem oder metastasierendem Endometriumkarzinom (Gebärmutterkrebs) erteilt hat, die ARID1A-Mutationen aufweisen und bei denen der Krebs nach mindestens einer vorangegangenen Behandlungslinie fortgeschritten ist. Die FDA vergibt den Fast Track-Status, um die Entwicklung und Prüfung von Wirkstoffen, die zur möglichen Behandlung schwerwiegender Erkrankungen oder Deckung eines ungedeckten medizinischen Bedarfs eingesetzt werden könnten, zu erleichtern und zu beschleunigen.

Tulmimetostat wurde entwickelt, um die EZH2-Inhibitoren der ersten Generation durch eine höhere Wirksamkeit, eine längere Verweildauer am Zielort und eine längere Halbwertszeit zu verbessern, was das Potenzial für eine verstärkte Anti-Tumor-Aktivität bietet. Der Fast Track-Status für Endometriumkarzinom wurde auf der Grundlage präklinischer Ergebnisse und vorläufiger klinischer Daten aus einer laufenden Phase-1/2-Studie erteilt. In dieser Studie wird Tulmimetostat als Monotherapie bei Patienten mit fortgeschrittenen soliden Tumoren oder Lymphomen untersucht, darunter ARID1A-mutiertes Endometriumkarzinom und klarzelliges Ovarialkarzinom, diffuses großzelliges B-Zell-Lymphom, peripheres T-Zell-Lymphom, BAP1-mutiertes Mesotheliom und kastrationsresistenter Prostatakrebs. Die aktualisierten Ergebnisse wurden auf der Jahrestagung 2023 der American Society of Clinical Oncology (ASCO) im Juni vorgestellt.

Tulmimetostat ist das dritte klinische Programm von MorphoSys, das von der FDA den Fast Track Status erhalten hat. Pelabresib, ein BET-Inhibitor, erhielt 2018 den Fast Track-Status für Myelofibrose, und Tafasitamab, eine auf CD19 abzielende Immuntherapie, erhielt diesen Status 2014 für das rezidivierte oder refraktäre diffuse großzellige B-Zell-Lymphom.

Über Tulmimetostat
Tulmimetostat (CPI-0209) ist ein in der Entwicklung befindliches Prüfpräparat, das die Antitumoraktivität fördern soll, indem es die Funktion der Proteine Enhancer of Zeste Homolog 1 und 2 (EZH2 und EZH1) hemmt um stillgelegte Gene wie Tumorexpressorgene zu reaktivieren. Tulmimetostat wird als eine einmal täglich oral zu verabreichende Behandlung in einer Phase-1/2-Studie (NCT04104776) bei Patientinnen und Patienten mit fortgeschrittenen soliden Tumoren oder Lymphomen, einschließlich ARID1A-mutiertes Klarzellkarzinom des Eierstocks und der Gebärmutter, diffuses großzelliges B-Zell-Lymphom, peripheres T-Zell-Lymphom, BAP1-mutiertes Mesotheliom und kastrationsresistentem Prostatakrebs untersucht. Zu den primären Zielen der Studie gehören die Bestimmung der maximal verträglichen Dosis und/oder der empfohlenen Phase 2-Dosis sowie die Bewertung der Antitumoraktivität der Tulmimetostat-Monotherapie. IZB-alumnus MorphoSys AG (FSE: MOR; NASDAQ: MOR) announced in mid-September that the U.S. Food and Drug Administration (FDA) granted Fast Track designation for tulmimetostat, the company’s investigational next-generation dual inhibitor of EZH2 and EZH1, for the treatment of patients with advanced, recurrent or metastatic endometrial cancer harboring AT-rich interacting domain containing protein 1A (ARID1A) mutations and who have progressed on at least one prior line of treatment. The FDA grants Fast Track designation to facilitate the development and expedite the review of medicines intended to treat serious conditions and potentially address an unmet medical need, with the goal of getting these important, new therapies to patients earlier.

Tulmimetostat was designed to improve on first generation EZH2 inhibitors through increased potency, longer residence time on target and a longer half-life, offering the potential for enhanced anti-tumor activity. The Fast Track designation in endometrial cancer was granted based on preclinical results and preliminary clinical data from an ongoing Phase 1/2 study. This study is investigating tulmimetostat as a monotherapy in patients with advanced solid tumors or lymphomas, including ARID1A-mutated endometrial carcinoma and ovarian clear cell carcinoma, diffuse large B-cell lymphoma, peripheral T-cell lymphoma, BAP1-mutated mesothelioma and castration-resistant prostate cancer. Updated results were presented at the 2023 American Society of Clinical Oncology (ASCO) Annual Meeting in June.

Tulmimetostat is MorphoSys’ third clinical program to receive Fast Track designation from the FDA. Pelabresib, an investigational BET inhibitor, received Fast Track designation for myelofibrosis in 2018, and tafasitamab, a CD19-targeting immunotherapy, received this designation for relapsed or refractory diffuse large B-cell lymphoma in 2014.

Tulmimetostat (CPI-0209) is an investigational compound designed to exert anti-tumor activity by inhibiting the function of enhancer of zeste homolog 1 and 2 (EZH2 and EZH1) proteins to reactivate silenced genes like tumor suppressor genes. Tulmimetostat is being tested as a once-daily oral treatment in a Phase 1/2 trial (NCT04104776) in patients with advanced solid tumors or lymphomas, including ARID1A-mutated ovarian clear cell carcinoma and endometrial carcinoma, diffuse large B-cell lymphoma, peripheral T-cell lymphoma, BAP1-mutated mesothelioma and castration-resistant prostate cancer.
